ASS
ASS (Advanced SubStation Alpha) is a subtitle script format edited in tools such as Aegisub. Besides timed dialogue it stores styles, fonts, colors, positions, karaoke and animation effects, which is why anime fansubs and many MKV releases use it.
SRT
SRT (SubRip) is the most widely supported subtitle format: a plain-text list of numbered subtitles, each with a start and end time. VLC, smart TVs, video editors and YouTube read it, and it supports only simple tags such as bold, italic and underline.

Advanced SubStation Alpha is the styled subtitle format of anime fansubs and Aegisub. An ASS script defines named styles with fonts, colors, outlines and shadows, and override tags place, animate and fade text anywhere on screen, including karaoke effects and typeset signs. VLC, mpv and MKV players render it.
TARGET
SubRip Text began with the SubRip ripping tool in the early 2000s. Each subtitle is a number, a start and end time with a comma before the milliseconds, and one or more lines of text. Premiere Pro, DaVinci Resolve, VLC, smart TVs, YouTube, Facebook and LinkedIn all accept SRT.

Karaoke-effect lines for openings and endings are removed, and the script's original karaoke line is used when it has one, so the lyrics appear once as plain text. A script without those original lines loses the song lyrics.
Drop embedded fonts from a script
Aegisub scripts that embed fonts in a [Fonts] section carry a lot of extra data. The SRT keeps only the dialogue and its timings.
Read scripts saved in old encodings
Scripts saved in UTF-16 or old code pages are detected and written as UTF-8 SRT, so accents and non-Latin text display properly.
Quote dialogue on a fan wiki
Wiki editors get readable lines with timings instead of a script full of style tags.

Without override codes, the SRT reads like a plain script, so typos and timing gaps are easy to spot.

TROUBLESHOOTING
Most issues come from features one format has and the other doesn't.

SRT can't show positions, fonts, colors or animation, so typesetting is removed.

Karaoke effect lines are removed. If the script has no original karaoke line, the song has no subtitles.

Bold that comes only from a style is a typeface choice in ASS, so it isn't marked.

Subtitle files over 50 MB are refused. Real subtitle files are far smaller.

The encoding is detected automatically. A very short file whose only accented letters fit several code pages can be misread; re-save it as UTF-8 and convert again.

Video files, picture-based subtitles and other binary files renamed as subtitles are refused with a message saying what they are.
